• 该试剂盒是否可以用于细菌样本的染色？

  不可以，因为该试剂盒的核染料是对完整细胞核进行染色，细菌不具有成形细胞核，所以不建议使用该试剂盒检测。

• 该试剂盒中的LiveDye和NucleiDye应如何保存？

  请于-20℃避光保存。

• 流式操作过程中设置了一个未作荧光染色的阴性对照组目的是什么？

  在流式实验中阴性对照的作用是调试流式细胞仪。如有疑问，请联系support@abbkine.com进行详细咨询。

• 该试剂盒是标记细胞的哪种指标？

  该试剂盒分别标记细胞的两种状态，红色荧光标记的是死细胞，绿色荧光标记的是活细胞。

• 同一个细胞上能同时染2个颜色，是怎么回事？

  如果细胞染色过程中，细胞培养条件没有保持好，活细胞染料进入细胞后，细胞趋近于死亡，细胞膜缺损，导致PI进入细胞，将细胞核着色。

• 做脱落细胞的活死细胞染色，胰酶消化会不会对染色有影响？

  可以，需要进行流式仪计数，显微镜无法对脱离细胞进行观察，胰酶消化对细胞活力一般不会有影响，除非客户的细胞比较敏感

• 试剂盒内试剂温度应该如何控制？

  建议现配现用，而且母液要一直放在-20度保存，在使用之前配成工作液并37度水浴。

• 该试剂盒能否用于水凝胶培养的细胞染色？

  采用钙黄绿素-AM (Calcein-AM) 和碘化丙啶 (PI) 试剂对细胞进行荧光染色，其中绿色荧光指示活细胞，红色荧光指示死细胞。指示活细胞的绿色荧光细胞团在水凝胶中均匀分布，指示死细胞的红色荧光基本观察不到，表明3D细胞培养水凝胶对胎儿骨髓间充质干细胞的细胞毒性很低，拥有良好的细胞相容性。

• 如果样本用DMSO做保护剂，是否对该试剂盒的结果有干扰？

  可以，因为我们这个试剂盒里面的染料助溶剂是DMSO，样品中的DMSO不会对试剂盒的作用有影响。

• 该试剂盒如何评价细胞存活率？

  用活细胞个数比上总细胞个数。
